Extensive transcription mis-regulation and membrane defects in AdipoR2-deficient cells challenged with saturated fatty acids

چکیده
How cells maintain vital membrane lipid homeostasis while obtaining most of their constituent fatty acids from a varied diet remains largely unknown. Here, we used transcriptomics, lipidomics, growth and respiration assays, property analyses in human HEK293 or umbilical vein endothelial (HUVEC) to show that the function AdipoR2 is respond rigidification by regulating many metabolism genes. We also AdipoR2-dependent critical for challenged with saturated acids. Additionally, found deficiency causes transcriptome cell physiological defects similar those observed SREBP-deficient upon SFA challenge. Finally, compared several genes considered important homeostasis, namely AdipoR2, SCD, FADS2, PEMT ACSL4, SCD are among these prevent excess saturation when exogenous SFAs. conclude one primary mechanisms protects against

The adiponectin receptor AdipoR2 and its Caenorhabditis elegans homolog PAQR-2 prevent membrane rigidification by exogenous saturated fatty acids
Dietary fatty acids can be incorporated directly into phospholipids. This poses a specific challenge to cellular membranes since their composition, hence properties, could greatly vary with different diets. That vast variations in diets are tolerated therefore implies the existence of regulatory mechanisms that monitor and regulate membrane compositions. Here we show that the adiponectin recept...
متن کاملRegulation of gene transcription by fatty acids.
Dietary fat is well recognised as an important macronutrient that has major effects on growth, development and health of all animals including humans. The amount and type of fat in the diet impacts on many aspects of metabolism including lipoprotein pathways, lipid synthesis and oxidation, adipocyte differentiation and cholesterol metabolism. It has become increasingly apparent that many of the...
متن کاملCompared with saturated fatty acids, dietary monounsaturated fatty acids and carbohydrates increase atherosclerosis and VLDL cholesterol levels in LDL receptor-deficient, but not apolipoprotein E-deficient, mice.
Heart-healthy dietary recommendations include decreasing the intake of saturated fatty acids (SFA). However, the relative benefit of replacing SFA with monounsaturated fatty acids (MUFA), polyunsaturated fatty acids (PUFA), or carbohydrates (CARB) is still being debated. We have used two mouse models of atherosclerosis, low density lipoprotein receptor-deficient (LDLRKO) and apolipoprotein E-de...
متن کاملFatty Acids: Straight-chain Saturated
Straightor normal-chain, saturated components (even-numbered) make up 10-40% of the total fatty acids in most natural lipids. The most abundant saturated fatty acids in animal and plant tissues are straight-chain compounds with 14, 16 and 18 carbon atoms, but all the possible oddand even-numbered homologues with 2 to 36 carbon atoms have been found in nature in esterified form. They are named s...
متن کاملExtensive impact of saturated fatty acids on metabolic and cardiovascular profile in rats with diet-induced obesity: a canonical analysis
BACKGROUND Although hypercaloric interventions are associated with nutritional, endocrine, metabolic, and cardiovascular disorders in obesity experiments, a rational distinction between the effects of excess adiposity and the individual roles of dietary macronutrients in relation to these disturbances has not previously been studied. This investigation analyzed the correlation between ingested ...
